Make doubling dilutions of standard from 1-1 to 10-1Use blood as diluent…Quantity used 200ul of standard and 200ul of blood.
Spot 30µL of the diluted standard series onto each of 5 spots on the card (do not spot the neat standard)
Allow to dry for at least 24 hours at Room temperature
Store card (with dessicant)
Prior to testing:
Cut out individual spots
Elute with 330 ml PBS/Tween 0.05% into false bottom tubes
PBS/Tween was prepared by adding 10ml of PBS buffer + 5ul of tween to get 0.05% of tween.
Store eluates at 4oC

Specimens: 50 patients samples.
Make 1:2 dilution of aliquot serum in blood as diluent
Spot 30µL of diluted samples onto spots on filter paper cards
Air dry for at least 24 hours at room Temperature
Store cards (with dessicant)
Prior to testing:
Cut out individual spots
Elute with 330 ml PBS/Tween 0.05% into false bottom tubes
Same preparation as used in the standards
Store eluates at 4oC

Make the negative controls, neat blood which was diluted with buffer.
And 20 spots were obtained.. then same procedure was carried out.

write how this test is performing what are the reagents used? What are the important instructions to do during the test? What is the principle of the test? Cover
everything writing on the attached file from the Machine’s company.

You must find new scientific reference for the methodology except the final step which was done by cobas machine because I added the reference for it.
You must find reference for Dried Blood Spots preparation and PBS/Tween buffer and everything used. Also, write all the materials used.
The reference must not be more than 5 yrs old. The work much be cited inside text. Reference must be done in Harvard.
